THE DEVELOPMENT OF AUTOMATED TOOLS FOR COUNTING THE NUMBER OF MICROORGANISMS ON SOLID NUTRIENT MEDIA

ABSTRACT

The purpose of this study was to develop software for the determination of the area occupied by colonies on a petri dish on digital photos.
The developed program can be useful in various studies related to counting the number of microorganisms in environmental, to assess the survival of bacteria under the influence of different factors.
The program «PetriEye» written in Java and can be used on all platforms and operating systems for which there is JVM.
The effectiveness of the programs was tested in experiments of the photodynamic action on Sthaphylococcus aureus 209 P in vitro. Effect on the bacteria was carried out by LED (405 nm, 5.31 mW/cm2) for 5, 10, 15 and 30 minutes. The number of colony forming units (CFU) was determined by counting the direct vision, using the «PetriEye» defined area (S), occupied by the colonies on a petri dish.
Shown that the values of the relative density of the bacterial population obtained by the two methods have differences within tolerance. The values obtained using the «PetriEye» generally 2-4% higher than the direct counting. Thus, the use of the program allows to eliminate the human factor, inevitable under direct visual counting, speed up the processing of the experimental results.
